RIGOXXON red is a synthetic double stranded RNA (dsRNA) with a 5´-triphoshate (5´-ppp). It has a length of 20 bp. It is composed of cytosines and guanosines. It is linked to a red fluorophore (Abs./Em.=555/575 nm).
Double stranded RNA with a 5´-ppp is a potent activator of innate immunity. In the context of innate immunity, 5´-ppp-dsRNA is a pathogen associated molecular pattern (PAMP) that activates innate immune response through pathogen recognition receptors (PRR). The PRR of 5´-ppp-dsRNA is the retinoic-acid inducible gene I (RIG-I) protein [1,2].
RIG-I is present in the cytosol of antigen presenting cells such as dendritic cells (DCs) and monocytes [3]. Signaling of RIG-I is triggered by a dsRNA bearing a 5´-triphosphate (5´-ppp) [3].
Triggering the RIG-I-pathway IFN-α and IFN-β production, improves cross-presentation of antigens and MHC class I expression.
In animal models, 5´-ppp-dsRNA enhances the specific T cell responses. Moreover, RIGOXXON red displays a resistance to serum and body fluids. It is therefore ideal for experimental set-up in cell culture supplemented with Fetal Calf Serum (FCS) and in small animal models.
